چکیده

The application of voltammetric methods using different rates of polarisation on HMDE reveal inert or labile behaviour of Cd- or Zn- complexes in the presence of excessive cadmium or zinc ions in solution. This phenomenon was demonstrated first on the simplest phytochelatin - complex of peptide (gamma-Glu-Cys)(2) Gly with cadmium, later on rabbit liver metallothioneins - Cd(7) MT in the presence of cadmium and Cd(5) Zn(2) MT in the presence of zinc. Voltammetric methods can distinguish between labile and inert complexes present simultaneously and therefore could elucidate their role in reactions of metal ion transfer.Another method using different rates of polarisation - elimination voltammetry with linear scan - proved that S-tetracoordinated complexes of Cd(II) or Zn(II) in the above-mentioned metallothioneins on HMDE are reduced in the adsorbed state. This implies the possibility of increasing the sensitivity of identification or determination of the above complexes. On carbon composite electrode, similar behaviour of Cd-complexes as on HMDE was observed using differential pulse voltammetry.

Voltammetric investigation of surface-confined proteins.

Development of sensitive and selective methods to detect proteins at trace levels is of great biological importance. Via derivatization with a bifunctional cross-linker 4-maleimidobutyric acid N-hydroxysuccinimide ester (GMBS) and an electrochemical marker 11-ferrocenyl-1-undecanethiol (Fc-SH), voltammetric determination of surface-confined proteins electrostatically adsorbed onto the polyelect...

Amplified voltammetric detection of glycoproteins using 4-mercaptophenylboronic acid/biotin-modified multifunctional gold nanoparticles as labels

Ultrasensitive detection of protein biomarkers is essential for early diagnosis and therapy of many diseases. Glycoproteins, differing from other types of proteins, contain carbohydrate moieties in the oligosaccharide chains. Boronic acid can form boronate ester covalent bonds with diol-containing species. Herein, we present a sensitive and cost-effective electrochemical method for glycoprotein...

Quantification of metals released by metallothionein adsorbates at mercury film electrodes by electrochemical ICP-atomic emission spectrometry

Electrochemistry combined on-line with inductively coupled plasma-atomic emission spectrometry (EC/ICP-AES) was used to study the electrode reactions of rabbit liver metallothionein (MT) adsorbates at thin mercury films (TMFs). Three major oxidation peaks, with peak potentials at −1.20, −0.75 and −0.32 V, were observed. Through the correlation of the DPV characteristics to the elution of the Zn...

The Effect of Cadmium on the Ultrastructure and Metallothionein Levels in the Liver and Kidneys of Japanese quail

Background: The aim of this study was to use Japanese quail as an animal model to evaluate the effects of cadmium (Cd) on the ultrastructure and the activity of metallothionein (MT) in the liver and kidneys. Methods: One hundred male Japanese quails were randomly divided into two Cd and control groups in 2015. The first group received 100 ppm Cd for 60 days in their feed. The ultrastructural c...

Catalase and Metallothionein genes expression analysis in wheat cultivars under drought stress condition

Drought stress is one of the serious problems that restricted agronomic plant production worldwide. In molecular level, the harmful effect of drought stress is mostly caused by producing of large amount of reactive oxygen species (ROS). Catalase and Metallothionein genes have a crucial role to mope the hydrogen peroxide (H2O2) resulting reducing oxidative damage. In this research the gene expre...
